Clinical utility

Bone marrow collection for hematopoietic progenitor cell transplantation (HPCT) contains a high number of red blood cells (hematocrit around 30-35%) and plasma.

In some situations, such as donor/recipient ABO incompatibility, excess volume greater than 20 ml/kg/patient, cryopreservation or history of allergic reactions, it is necessary to reduce the number of red cells and plasma and recover the fraction of nucleated cells (buffy coat) from bone marrow where the hematopoietic progenitor cells (HPCs) are found.

Method

Automated bone marrow (BM) buffy coat processing is faster and less operator dependent compared to manual processing. Moreover, since it is carried out in a closed system, the risk of contamination is lower.

The Banc de Sang i Teixits (Blood and Tissue Bank, BST) uses the Sepax system, which consists of a centrifuge system and disposable equipment. The centrifuge allows the separation of BM blood into different components using an optical light signal system. Following several repeat cycles depending on the initial volume, the device delivers the components in separate bags (buffy coat, red blood cells, plasma).

Reference values

The expected recovery of nucleated cells is > 75%.
